If you've on a BSD system, you have a vacation program already. Otherwise, you might want to get the Deliver program, written by yours truly. It runs shell script(s) at message delivery time; those shell scripts can do just about anything you want, including send vacation notices. In fact, Deliver 2.0 (STILL in the c.s.u queue!) includes a complete vacation notice script as a sample.
