Stephen, no you don't need to use the key to open that door into the Dain
Corbum room...  either the key or button, doesn't matter which..
 
The "key" to getting to the Corbum is the inscription on the wall near
the Corbum...  not everything in this Dungeon is actually as it appears
to be.  Close your eyes and "use the Force"... or, drop unneeded items..
 
Regarding the Skeleton Key lock from ROS... you don't need to open that
door... you'll find your way to the other side eventually anyway..
the way to the ROS corbum is through the DDD...
