A colleague of mine is using GUS as a reporter gene for promoter
action in a parasitic fungus, and using X-GLUC for histochemical assay.
He doesn't seem to be getting very good signal strength, despite using
a "very strong" promoter. Does anyone have any tips for optimizing
GUS/X-GLUC histochemistry, especially in fungal hyphae?
