DigiBoard ? Anvil (sp ??) ? or something else ? The ability to accomodate more than one board seems like a necessity. Okay, may be not 256 ports (I am pretty sure I saw an ad for 256 ports in one PC before), but how about 64 ports ? Full documentation on programming is of course very important.
