The phenotype is expressed in callus tissue and would aqppear to be a reasonable candidate for cloning by complementation using a library in a binary vector constructed either from YACs in the region of the gene or DNA of the appropriate size identified using nearby RFLPs and purified from a CHEF gel. Has anyone tried this approach for another selectable gene? Can anyone suggest a useful vector, preferably not subject to proprietary rights, for this approach?
