In that sense, it has flown. However, it is not operational, and has never been in space. It is used a lot for pathfinder tests. I.e., after you build a new piece of hardware that interfaces with the shuttle, you use the Enterprise to check it all out before committing to an operational shuttle. Although I have not seen it, I would guess that the AF ad was showing the Enterprise being used to test out the new military shuttle launch facility at Vandenberg AFB, CA.
