There's a switch on the LA50 (switch SW1, position 6) that controls whether XON/XOF or Ready/Busy is used; XON/XOF is selected by having the switch in the open position (towards the front of the printer). Something else to be aware of is that a very common failure mode of the LA50 PCB is that it loses XON/XOF synchronization (I suspect the buffer full check fails). I've seen this happen on at least two different printers. You should try borrowing someone else's LA50 and see if the problem goes away.
