Lewis Chris, I'm not sure this will help with your problem, but there is a fairly effective method of making progressive deletions in DNA (Henikoff. 1984. Gene 28:351). Modified T7 DNA polymerase (Tabor and Richardson. 1987. PNAS 84:4767) seems to solve a number of problems in sequencing. It is available as Sequenase from U.S. Biochemical Corp. in Cleveland. They mail out a flyer with several short technical reports on the use of this enzyme for sequencing. I am in no way affiliated with U.S. Biochemicals. I hope this is helpful. Bob -------
