I've used both 68010s and 68020s in Amigas since 1985, and several weeks ago operated a 68030 on the same machine. There are commercially available cards facilitating such conversion, with provision for 32-bit-wide RAM (static) cards attached "locally". Software changes require enabling the cache(s) and cognizance of the different stack frames. Operating systems such as that on the Amiga handle the different processors with ease, and Motorola publishes AppNotes detailing these steps in the general sense.
