Does anybody know whether larger fragments than the usual 20- to 30-mers of oligonucleotides can be used as primers for the PCR ? (My aim is to connect a certain marker-gene (ca. 1500 bp) instead of a restriction-site directly to the PCR-fragments to avoid cloning.) Which restriction enzymes do cut single-stranded DNA ? (HindIII works).
